Genetic Toxicity Testing

Peony pollen products need to be shown to be free from genotoxic risk, and Aprofood's toxicology team provides the data to demonstrate it. Our genetic toxicity studies cover raw pollen, wall-broken pollen, extracts and finished products, and return reliable mutagenicity and clastogenicity results with a clear interpretation. Product developers can rely on these findings for new ingredient development, registration and market entry.

Genetic Toxicity

Fig 1. Genetic Toxicity Testing

Genetic toxicity describes the capacity of a substance to damage genetic material, either through gene mutations (mutagenicity) or through structural or numerical chromosome damage (clastogenicity and aneugenicity). Because such damage can lead to serious long-term health effects, genetic toxicity testing is a core element in the safety evaluation of every food ingredient. A standard battery of in vitro and in vivo assays is applied, since no single test detects every type of damage. It covers gene mutation in bacteria, chromosome damage in mammalian cells and chromosome damage in living animals. Peony pollen has long been used as a food and traditional health material, yet wall-broken pollen, extracts and concentrated products differ in composition and contain higher levels of active compounds. Regulators therefore still ask for documented genotoxicity data on these products, because history of use alone cannot establish their safety, and a standardized battery is needed to rule out genotoxic risk for consumers.

Our Service

Aprofood's scientists bring extensive experience in toxicological safety evaluation to meet industry standards and regulatory requirements. Every study follows internationally recognized guidelines, including the OECD test guidelines and national food safety standards, and is tailored to the test material, its intended use and the target market. The following approaches are available for evaluating the genetic toxicity of peony pollen products.

Test Method Description
Bacterial Reverse Mutation Test (Ames Test)
  • Guideline: OECD 471
  • Test system: several Salmonella typhimurium strains and Escherichia coli WP2, with and without metabolic activation (S9)
  • Outcome: detects gene mutations (base-pair substitution and frameshift)
  • Application: first-line screening of all peony pollen products
In Vitro Mammalian Cell Micronucleus Test
  • Guideline: OECD 487
  • Test system: mammalian cells treated with and without S9
  • Outcome: detects chromosome breakage (clastogenicity) and chromosome loss (aneugenicity)
  • Application: assessment of chromosome damage in mammalian cells
In Vitro Chromosomal Aberration Test
  • Guideline: OECD 473
  • Test system: cultured mammalian cells, with and without S9
  • Outcome: detects structural chromosome aberrations
  • Application: clastogenicity evaluation of extracts and concentrated products
In Vitro Mammalian Cell Gene Mutation Test
  • Guideline: OECD 476 / 490
  • Test system: mouse lymphoma or other mammalian cell lines
  • Outcome: detects gene mutations and some chromosome-level events
  • Application: complementary evaluation of mutagenicity in mammalian cells
In Vivo Mammalian Erythrocyte Micronucleus Test
  • Guideline: OECD 474
  • Test system: rodent bone marrow or peripheral blood, oral dosing at several dose levels
  • Outcome: detects chromosome damage in a whole-animal system
  • Application: confirmation of in vitro results under real exposure conditions
Mammalian Spermatogonial Chromosomal Aberration Test
  • Guideline: OECD 483
  • Test system: rodent germ cells after oral dosing
  • Outcome: detects chromosome damage in germ cells
  • Application: additional evaluation when germ cell effects must be assessed

Our Applications of Genetic Toxicity Testing

Genetic toxicity data play a role at several stages of peony pollen product development and market entry. Aprofood designs each study to match the purpose behind it, as shown in the applications below.

Application Description
Pollen Genotoxicity Evaluation Raw and wall-broken pollen can be assessed for potential genotoxic effects, helping confirm the safety profile of pollen materials with a history of food use.
Extract Assessment Extracts and concentrated pollen ingredients may require separate evaluation when processing alters their composition or changes the levels of active compounds.
Novel Food Registration Genotoxicity data can form part of a novel food safety dossier, alongside acute, teratogenicity, and 90-day subchronic studies planned for the target market.
Finished Product Verification Marketed forms such as tablets, capsules, powders, and beverages can be tested using sample preparation procedures suited to each assay system.
Formulation Screening Different batches, processing methods, or formulations can be compared for genotoxic potential to support selection of the most suitable option before further development.

Our Workflow of Genetic Toxicity Testing

01
Study Design

We review the test material, intended use, target market, and study objectives, then develop a study protocol and quotation for confirmation. The protocol specifies dose levels, animal numbers, study duration, and applicable guidelines.

02
Sample Preparation

The test material is characterized and prepared into a suitable dosing formulation. Homogeneity and stability are assessed to ensure consistent and representative dosing throughout the study.

03
Animal Acclimation

Healthy animals are acclimated under controlled laboratory conditions before dosing. Temperature, humidity, lighting, diet, housing, and general health are monitored throughout the acclimation period.

04
Dosing & Observation

Animals receive the test material by oral gavage at the specified dose levels and are monitored throughout the study period. Clinical signs, behavior, food intake, body weight, and other relevant observations are recorded at scheduled intervals.

05
Necropsy & Data Analysis

Gross necropsy is performed at study completion, with histopathological evaluation conducted when required. Study data are analyzed to identify treatment-related findings and assess overall toxicity.

06
Final Reporting

A comprehensive report summarizes study procedures, observations, toxicity findings, and conclusions. Raw data and recommendations for further safety or toxicological studies are provided where applicable.

Advantages of Our Services

Peony-specific experience

We know how wall-breaking and extraction change the composition of peony pollen products, and design dosing and test plans accordingly.

Guideline-compliant studies

Studies follow OECD guidelines and national food safety standards, so the data suit regulatory submissions in different markets.

Reduced animal use

Limit tests and stepwise procedures are selected where appropriate, meeting ethical requirements while saving time and cost.

Connected safety program

Results link directly to genetic toxicity, reproductive toxicity and 90-day subchronic studies, so a complete safety evaluation can be planned in one project.

Experienced toxicology team

Our scientists have extensive experience in study design, data interpretation and safety assessment, and can explain the results clearly in the context of your product and market.

Clear and complete reporting
