Acute Oral Toxicity Study

Aprofood's toxicology team helps ensure that peony pollen products are safe for consumption. Our acute oral toxicity studies cover raw pollen, wall-broken pollen, extracts and finished products, and deliver reliable toxicity data together with a clear hazard classification. These results provide a solid basis for new ingredient development, product registration and market entry.

Acute Oral Toxicity

Fig 1. Acute Oral Toxicity

Acute oral toxicity refers to the adverse effects that appear within a short period, usually 14 days, after a single oral dose, or multiple doses given within 24 hours, of a test substance. It is the first step in the safety evaluation of any food ingredient, and its results are used to estimate the median lethal dose (LD50), observe signs of toxicity, identify target organs and select dose levels for later studies such as genetic toxicity and 90-day subchronic toxicity tests. Peony pollen has a long history of use as a food and traditional health material. However, modern markets still require documented safety data, especially for wall-broken pollen, extracts and concentrated products, whose composition and bioavailability differ from those of raw pollen. Such products cannot be assumed safe from history of use alone, so a standardized toxicity study is necessary to confirm that they do not pose a risk to consumers.

Our Service

The scientists at Aprofood have extensive experience in toxicological safety evaluation that meets industry standards and regulatory requirements. Studies are carried out in standardized animal facilities in accordance with internationally recognized guidelines, such as the OECD test guidelines, and each study is designed around the type of test material, its intended use and the requirements of the target market. We can determine the toxicity of peony pollen products through the following approaches.

Test Method Description
Limit Test
  • Guideline: OECD 420 / 423 (limit dose option)
  • Dosing: a single high dose (2000 or 5000 mg/kg body weight) given to a small group of animals
  • Outcome: confirms low toxicity without LD50 calculation
  • Application: food-grade pollen and products expected to be practically non-toxic
Fixed Dose Procedure
  • Guideline: OECD 420
  • Dosing: stepwise fixed doses (5, 50, 300 and 2000 mg/kg), usually 5 animals per step
  • Outcome: hazard classification based on the dose causing evident toxicity
  • Application: extracts and concentrated products with unknown toxicity
Acute Toxic Class Method
  • Guideline: OECD 423
  • Dosing: stepwise doses with usually 3 animals per step, depending on mortality
  • Outcome: hazard classification and estimated LD50 range
  • Application: screening and classification of test materials
Up-and-Down Procedure
  • Guideline: OECD 425
  • Dosing: one animal at a time, with the dose adjusted by the previous outcome
  • Outcome: LD50 estimate with confidence interval
  • Application: products that need a quantitative LD50 value
Clinical Observation & Body Weight Monitoring
  • Scope: applies to all methods
  • Procedure: daily observation for 14 days after dosing
  • Records: clinical signs, behavior, food intake, mortality and body weight
  • Outcome: onset, severity and recovery of toxic effects
Gross Necropsy & Histopathology
  • Scope: applies to all methods
  • Procedure: necropsy and organ weights at the end of the observation period
  • Histopathology: performed on selected tissues when abnormalities are found
  • Outcome: target organs and nature of tissue damage

Our Applications of Acute Oral Toxicity Study

Acute oral toxicity data are used at several stages of peony pollen product development and market entry. Aprofood supports the following applications with study designs matched to each purpose.

Application Description
Pollen Safety Evaluation Safety confirmation for food-grade pollen with a long history of use, toxicity classification for raw and wall-broken pollen, and dose-range data for later safety studies.
Extract Assessment Evaluation of extracts whose composition differs from raw pollen, identification of the dose range that causes evident toxicity, and support for safe use level decisions during formulation.
Novel Food Registration Acute toxicity data required in safety dossiers, which can be planned together with genetic toxicity, teratogenicity and 90-day subchronic studies around the regulatory pathway of your target market.
Finished Product Verification Testing of tablets, capsules, powders and beverages as marketed, with the dosing formulation adapted to solubility and particle size, to confirm that the final formulation poses no acute risk.
Formulation Screening Comparison of the acute toxicity of different batches, processes or formulations, helping to select the safest option before further investment and guiding the design of later studies.

Our Workflow of Acute Oral Toxicity Study

01
Study Design

We review the test material, intended use, target market, and study objectives, then develop a study protocol and quotation for confirmation. The protocol specifies dose levels, animal numbers, study duration, and applicable guidelines.

02
Sample Preparation

The test material is characterized and prepared into a suitable dosing formulation. Homogeneity and stability are assessed to ensure consistent and representative dosing throughout the study.

03
Animal Acclimation

Healthy animals are acclimated under controlled laboratory conditions before dosing. Temperature, humidity, lighting, diet, housing, and general health are monitored throughout the acclimation period.

04
Dosing & Observation

Animals receive the test material by oral gavage at the specified dose levels and are monitored throughout the study period. Clinical signs, behavior, food intake, body weight, and other relevant observations are recorded at scheduled intervals.

05
Necropsy & Data Analysis

Gross necropsy is performed at study completion, with histopathological evaluation conducted when required. Study data are analyzed to identify treatment-related findings and assess overall toxicity.

06
Final Reporting

A comprehensive report summarizes study procedures, observations, toxicity findings, and conclusions. Raw data and recommendations for further safety or toxicological studies are provided where applicable.

Advantages of Our Services

Peony-specific experience

We know how wall-breaking and extraction change the composition of peony pollen products, and design dosing and test plans accordingly.

Guideline-compliant studies

Studies follow OECD guidelines and national food safety standards, so the data suit regulatory submissions in different markets.

Reduced animal use

Limit tests and stepwise procedures are selected where appropriate, meeting ethical requirements while saving time and cost.

Connected safety program

Results link directly to genetic toxicity, reproductive toxicity and 90-day subchronic studies, so a complete safety evaluation can be planned in one project.

Experienced toxicology team

Our scientists have extensive experience in study design, data interpretation and safety assessment, and can explain the results clearly in the context of your product and market.

Clear and complete reporting

Each report includes the protocol, raw data, observations and conclusions, giving you well-documented results that are ready for technical files and regulatory dossiers.
